研究概要	Gタンパクに共役する7回膜貫通型受容体のうちリガンドが不明なオーファン受容体の内因性リガンドとして近年発見されたペプチドのストレス反応への関与について検討した。具体的には、オレキシン、ニューロメジンUおよびガラニン様ペプチド(GALP)に着目して、主に視床下部-下垂体系への影響を検討した。その結果、以下の点について明らかにした。 (1)オレキシンは視床下部外側野にその産生ニューロンが限局して存在する。これらのニューロンは、拘束ストレスおよび寒冷ストレスにより興奮することを明らかにした。また、ラット脳室内に投与したオレキシンは視床下部室傍核と扁桃中心核に存在するCRFニューロンを選択的に興奮させることを明らかにした。 (2)ニユーロメジンUのラット脳室内投与により視床下部室傍核に存在するCRFニューロンを興奮させることを明らかにした。なお、扁桃中心核のCRFニューロンは興奮させなかった。 (3)GALPは、下垂体後葉では後葉細胞で産生される。この産生は、浸透圧刺激、菌体毒素投与によるエンドトキシンショックおよび慢性炎症刺激により増加することを明らかにした。また、遺伝性肥満ラット(fa/fa)でも増加しており、レプチン非依存性の機序も存在することを明らかにした.後葉細胞が産生・分泌したGALPは下垂体後葉からのバゾプレッシン分泌を引き起こすことも明らかにした。GALPは弓状核ニューロンでも産生されており、浸透圧刺激では変化しないこと、エンドトキシンショックで増加することを見出した。新規に発見された摂食関連ペプチドであるオレキシン、ニューロメジンUおよびGALPが、ストレスに対する内分泌反応の中軸をなす視床下部-下垂体-副腎系を賦活すること、下垂体後葉系を修飾することを明らかにした。
