研究概要

イヌ動脈瘤モデルの作製および解析
1.イヌ大動脈(頚動脈)瘤モデルの経時的解析
方法:ビーグル犬を麻酔下(ペントバルビタール35mg/kg、i.v.)、頚動脈内にエラスターゼを2時間浸透させた。術後1、2週の時点で、麻酔下、頚動脈の直径をエコーにて測定し、その後、摘出した血管を用いて以下の解析を行った。
結果:エラスターゼ作用後1週より頚動脈の直径は有意に増加し、摘出した血管では、キマーゼ活性が顕著に増加していた。また、組織のアンジオテンシンII産生活性も顕著に増加していた。
エラスターゼ作用後2週では、さらに頚動脈直径、キマーゼ活性およびアンジオテンシンII産生活性は増加する傾向であった。組織学的解析結果より、キマーゼの発現細胞は、外膜側に多く発現していることを確認した。
2.イヌ腹部大動脈瘤モデルの経時的解析
方法:ビーグル犬を麻酔下(ペントバルビタール35mg/kg、i.v.)、腹部大動脈内にエラスターゼを2時間浸透させる。術後1、2、4、8週の時点で、麻酔下、腹部大動脈の直径をエコーにて測定し、その後、摘出した血管を用いて以下の解析を行った。
結果:エラスターゼ作用後1週より腹部大動脈の直径は有意に増加し、摘出した血管では、キマーゼ活性が顕著に増加していた。また、組織のアンジオテンシンII産生活性も顕著に増加していた。エラスターゼ作用後2、4、8週では、経時的に頚動脈直径、キマーゼ活性およびアンジオテンシンII産生活性は増加する傾向であった。
以上、2つの大動脈瘤モデルにおいて、キマーゼ依存性アンジオテンシンII産生の亢進に伴い、大動脈瘤経が進展することが明らかになった。現在、イヌ腹部大動脈瘤モデルにおいて、キマーゼ阻害薬の大動脈瘤経に対する効果を検討しているところである。
