転写因子群による血液細胞・分化の制御機構の解明

研究概要

本研究の目的は、血球系転写因子群の血液細胞の分化における機能を明らかにすることである。現在、血液細胞を含む臓器の分化は、複数の転写因子の幹細胞内でのバランスで決定されるとの考え方が有力であるが、その仮説を個体内で検証する。血球系転写因子群の機能解析を通して、転写因子による血液細胞の分化の方向性の制御機構に、分子レベル・個体レベルで詳細にアプローチする。
a)MafB遺伝子欠損マウスの解析:これまでの細胞培養系を用いた実験より、単球系細胞の分化にはc-Maf、MafB等の大Maf転写因子群が必須な機能を果たしていることが示唆されている。既に申請者らは、c-Maf遺伝子欠損マウスの作製を行いその表現型を解析した。c-Maf遺伝子欠損マウスは、目の水晶体線維細胞の分化および増殖不全が認められたが、単球系の細胞分化を含め、血液系の細胞には異常が認められなかった。この結果は、単球系にはc-Mafと同様にMafBが発現しており、MafBにより機能が相補された可能性と、個体内では、単球系の細胞分化には大Maf群転写因子が必須ではない可能性が考えられる。前者の可能性を検証するために、MafB欠損マウスを作製しその表現型を解析した。その結果、マクロファージの分化が、阻害されていることが明らかになった。
b)GATA転写因子群欠損マウスにおける好酸球分化め解析:これまでの我々の解析で、GATA-1は赤血球の赤芽球から成熟赤血球への分化段階で、また、巨核球から虫小板の放出に必須の転写因子であることが明かとなっている。一方、好酸球分化におけるGATA-1の機能は、マウスでは末梢血に存在する好酸球数が非常に少ないこと、好酸球の培養が、ヒトに比較して難しいこと等から、解析がほとんど行われていなかった。そこで、我々が作製したGATA-1ノックダウンマウスを用いて、好酸球の分化状態を解析したところGATA-1が欠損すると好酸球が形成されないことが明らかになった。
