生体防御を担う活性酸素生成型食細胞NADPHオキシダーゼの活性化の分子機構

研究概要	NADPHオキシダーゼは活性酸素を生成する酵素であり、生成された活性酸素は生体防御において極めて重要な役割を担う。活性酸素生成型NADPHオキシダーゼのプロトタイプは食細胞に豊富に存在することから食細胞NADPHオキシダーゼとも呼ばれるが、その酵素本体は膜蛋白質gp91^<phox>であり、p22^<phox>と会合してシトクロムb_<558>を形成している。本酵素の活性化には、特異的アダプター蛋白質(p47^<phox>,p67^<phox>,とp40^<phox>:各々SH3ドメインをもつ)が刺激依存性に細胞質から細胞膜に移行してシトクロムb_<558>と相互作用する必要がある。私共はオキシダーゼの活性化機構を研究し、平成14年度は以下のような成果を得た。 (1)p47p^<phox>のPXドメインのphosphoinositides結合能が、p47^<phox>の膜移行及びオキシダーゼ活性化に必須であることを明らかにし、更に、この脂質結合能はp47^<phox>のリン酸化により誘導されることを示した。 (2)もう1つのオキシダーゼ活性化に必須の蛋白質p67^<phox>はそのC末側SH3ドメインを用いてp47^<phox>と結合するが、この結合が新しい様式のものであることを示し、複合体の立体構造を決定した。 (3)p67^<phox>に恒常的に結合している蛋白質として知られていたp40^<phox>の役割は不明であったが、p40^<phox>はp67^<phox>に結合してp67p^<phox>(更にはp47^<phox>)の膜移行を促進することにより、オキシダーゼ活性化を正に制御していることを明らかにした。 (4)新規なオキシダーゼ調節蛋白質であるp41^<nox>(p47^<phox>のホモログ)及びp51^<nox>(p67^<phox>のホモログ)を同定・クローニングした。更に、これらがgp91^<phox>ばかりでなく新規オキシダーゼNox1を活性化できることを示した。
