研究概要	非ケトーシス型高グリシン血症・脳症はグリシン開裂酵素系のP-、T-、H-蛋白質と呼ばれる固有の構成蛋白質のいずれかの一次的障害に基づき、けいれん・知能障害などの中枢神経症状を示し最終的には予後不良の遺伝性疾患である。「グリシン脳症」における神経変性の基礎的メカニズムの解明は、現代医学における重要な課題となっている。本研究では中枢神経系を含む全身でH-蛋白質を欠損するノックアウトマウスを作製・解析することによって、神経変性非ケトーシス型高グリシン血症の成因や病態を明らかにする。平成14年度の研究は以下の通りに順調に進行し、期待されたような研究成果を得た。・マウス肝臓由来RNAを鋳型として用いたRT-PCR法により、マウスのグリシン開裂酵素系H-蛋白質遺伝子をコードするcDNA断片(300bp)を得た。・ES細胞由来高分子ゲノムDNAを用いてSau3A1消化ライブラリーを作製し、上記cDNA断片をプローブとしてスクリーニングを行った。スクリーニングの結果、15個の陽性クローンを得た。・LoxP間のゲノム遺伝子の切出しを行うためには、Creレコンビナーゼを細胞レベルで発現するマウスが必要である。そのため、受精卵を含む全ての細胞で働くPGKプロモーターの下流にCre遺伝子カセットを接続した遺伝子を持つトランスジェニックマウス(B)を、受精卵前核へのマイクロインジェクション法により作製した。・今後上記で得たH-蛋白質ゲノム遺伝子DNA断片をもとにして作製したノックインマウス(A)とPGK-Creトランスジェニックマウス(B)を交配して、グリシン開裂酵素系H-蛋白質遺伝子ノックアウトマウスを得る。
