研究概要

非ケトーシス型高グリシン血症・脳症はグリシン開裂酵素系のP-、T-、H-蛋白質と呼ばれる固有の構成蛋白質のいずれかの一次的障害に基づき、けいれん・知能障害などの中枢神経症状を示し最終的には予後不良の遺伝性疾患である。「グリシン脳症」における神経変性の基礎的メカニズムの解明は、現代医学における重要な課題となっている。本研究では中枢神経系を含む全進でH-蛋白質を欠損するノックアウトマウスを作製・解析することによって、神経変性非ケトーシス型高グリシン血症の成因や病態を明らかにする。平成15年度の研究は以下の通りに順調に進行し、当初期待されたような研究成果をこれまでに得た。
・前年度クローン化したマウスのグリシン開裂酵素系H-蛋白質遺伝子をコードするゲノムDNA断片(17kbp)を用いて相同組換え用ベクター(Hpro. cond. KO vector)を作製した。エキソン1周囲に2か所のloxP部位を導入し、Cre蛋白質の共存下でエキソン1切り出し→H-蛋白質欠損が起こるようにベクターを設計した。
・ES細胞培養系を用いて電気穿孔法によりES細胞にHpro. Cond. KO vectorを導入しネオマイシン耐性を指標としてスクリーニングを行った。1回目のスクリーニングの結果、2個の陽性クローンを、2回目のスクリーニングの結果、15個の陽性クローンを得た。
・得られた相同組換え体ES細胞株をC57Blackマウス由来胚盤胞にマイクロインジェクションして仮親に移植した。これまでにES細胞由来毛色80%以上のキメラマウス8匹を得た。キメラマウスの性別はオスが7匹、メスが1匹でありオスの割合が高かった。
・今後上記で得られたキメラマウスとCre蛋白質発現トランスジェニックマウスを交配して、グリシン開裂酵素系H-蛋白質遺伝子ヘテロ欠損マウスを得る。ヘテロ欠損マウス同士の交配により、ホモ欠損マゥウスを得たのち、さまざまな解析を行う。
