異常タンパク質の蓄積が引き起こす空胞形成及び細胞死の分子メカニズムの解析

研究概要	本研究では、Machado-Joseph病(MJD)発症の第1ステップを担うと考えられるMJD蛋白質を特異的にプロセシングする酵素を同定するために、MJD蛋白質が切断された細胞で薬剤耐住遺伝子が発現するシステムを構築し、MJD蛋白質を切断する活性の高いPC12細胞亜株を選別・樹立することに成功した。このPC12細胞亜株では、MJD蛋白質の切断活性が親株に比べて著しく亢進しており、MJD蛋白質は200〜250番目のアミノ酸の近傍で切断を受けると推測された。次に、これらの細胞株、すなわちMJDプロセッシング酵素活性のほとんど無い元の細胞とその活性が増強している細胞株から、mRNAを抽出し、DNAチップを用いた解析を行った。その結果、発現が増加、減少している遺伝子がそれぞれ数十個ずつ存在することが明らかになった。現在、これらの全長cDNAを単離して、機能解析を進めている。一方、MJD79蛋白質と相互作用する分子として同定したVCP内の二つのATPaseドメイン(D1,D2)の各変異体を一過的に細胞に発現させたところ、D2ドメインの変異体p97[K524A]が空胞化を伴う細胞死を誘導した。そこでp97[K524A]を安定に発現する神経細胞株(PC12)を樹立して表現型を詳しく調べた。その結果、D2ドメインがp97の主要なATPaseであること、p97のATPase活性の低下は小胞体の品質管理機構を乱し、結果として小胞体ストレスと小胞体の異常な膨潤を伴う細胞死を引き起こすことを見いだした。さらに核・膜画分にポリユビキチン化したタンパク質が異常に蓄積することを明らかにした。また、このストレスシグナルは、ASK1ノックアウト細胞で、大幅に減弱することを見いだした。
