フローアレイを用いた多数SNPs(1塩基多型)同時タイピングシステムの開発

研究概要	テーラーメイド医療やゲノム創薬に欠かせない情報としてSingle nucleotide polymorphisms(SNPs)が注目されており、その検出法も様々なものが考案されている。しかし、それらの多くは手技が煩雑であったり、高価であったり、特殊機器が必要であったりという問題点もある。我々は近年、注目されてきているフローアレイ技術をこのSNPs検出に応用できないかと研究を重ねてきた。本研究は複数のSNPsをhigh-throughputかつ簡便にsingle laser搭載のフローサイトメーターで解析しようとするものである。簡単に説明すると短い合成DNAを固定させた直径5μmから15μm程度のビーズを用いて、合成DNA中、ゲノムDNA中の1箇所のSNPsを検出することに成功している。具体的にはビーズ上に固定したオリゴヌクレオチドプローブと蛍光標識されたサンプルDNAをハイブリダイズさせる。このときサンプルDNAがビーズ上のプローブDNAと完全に相補的であれば、両者は結合し、ビーズが蛍光を発するが、相補的でなければビーズは蛍光を発さない。この蛍光量の差をフローサイトメーターで計測しSNPsのタイピングを行う。昨年度までに成功していた合成DNAによる3箇所のSNPs検出に加えて、今年度はゲノムDNAでも2箇所のSNPsタイピングに成功した。この成果は今年5月に開かれる第22回国際細胞解析学会(フランス)での発表を予定している。本研究においてフローサイトメーターをもちいたSNPタイピング法の基礎は確立できたと考えており、今後この方法をDNAのみならずタンパク解析などにも応用していきたい。
