チオレドキシントランスジェニックマウスの炎症・ストレス耐性の分子機構の解析

研究概要	チオレドキシン(TRX)はその活性部位に-Cys-Gly-Pro-Cys-という2つのシステインをもち、基質蛋白のdisulfide結合還元能を有する分子量12kDの蛋白である。TRXは種々の酸化ストレスにより誘導され、細胞外へも放出される。TRXを過剰発現させたトランスジェニックマウス(TRX Tg)は対照マウスと比較して、脳虚血や紫外線障害に抵抗性を示し長寿傾向を示す。TRX Tgは対照マウスと比較し、Bleomycinや炎症性サイトカインであるIL-2とIL-18の併用投与により発症する間質性肺炎モデルにおいて、肺間質への白血球浸潤や肺間質の浮腫を著明に抑制した。thioacetamide投与による急性肝障害モデルにおいても、TRX Tgは対照マウスと比較して肝細胞における酸化ストレス障害の軽減を認め、肝細胞のapoptosisを抑制し、急性肝障害の軽減を認めた。また腎虚血再潅流障害が抑制されることも明らかにした。さらに、遺伝子組換えTRX蛋白投与でも、マウスにおける、脳梗塞の軽減、間質性肺炎の軽減作用を認めた。マウスにおいて、血中に投与した遺伝子組換えTRX蛋白により、白血球の炎症部位への血管外漏出が抑制され、炎症を抑制できることを明らかにしたほか、細胞外から投与した遺伝子組換えTRX蛋白の部は細胞に取り込まれること、細胞外から投与したTRXにより細胞内の活性酸素の発生が抑制され、caspase-3の活性化およびapoptosisも抑制されることを明らかにし、遺伝子組換えTRX蛋白の投与は種々の炎症及び酸化ストレス疾患の治療に有効であることが示唆された。
