キラーT細胞誘導におけるケモカイン受容体CCR5の役割の確立とその分子機序の解析

研究概要	免疫シナプスに集積し、抗原認識の感受性を制御するケモカイン受容体CCR5/CXCR3 ケモカイン/ケモカイン受容体の新たな機能を見出すために、AIDSの共受容体の一つであるCCR5に焦点を絞り、特にT細胞受容体(TCR)刺激とのクロストークの解明とそれらの生理学的かつ病理学的意義を明らかにすることを目的とした。まずCCR5はTCR刺激に伴って、そのリガンドであるRANTESと共に免疫シナプスに集積することを見出した。そしてTCR刺激でCCR5が実際に活性化されることをBRET (Bioluminescence Resonance Energy Transfer)法を用いて明らかにした。またTCR刺激に伴うLFA-1の活性化には、CCR5の活性化に伴うRap1の活性化が重要であることを示した。そこでCCR5/CXCR3ダブル遺伝子欠損マウスを作製することにより、ケモカイン受容体CCR5とCXCR3がCD8+T細胞の増殖へのコミットメントに重要であることをin vivoで確認した。以上よりTCR刺激後に、免疫シナプス腔にRANTESなどのケモカインが分泌され濃縮されたRANTESは、免疫シナプスに集積してきたCCR5を活性化させるだけの有効な濃度を保持することが可能であり、これが少なくともLFA-1の活性制御を介してT細胞の抗原認識の感受性を制御し、TCR刺激におけるboosting signalあるいはsustained signalの候補の一つとしてシグナルの持続に貢献し、T細胞が増殖するかどうかのコミットメントに重要なステップであることが推測された。このLFA-1を介したメカニズムが臓器移植拒絶反応の制御を説明する分子基盤を与えるものであり、また最近報告されたヒト腎臓移植におけるCCR5delta32をもつレシピエントの長期予後の改善の理論的根拠の一つを与えるものである。
