二酸化窒素様活性窒素種の心血管系組織からの発生機序とその制御に関する研究

研究概要	ラット心臓に、ペルオキシダーゼ活性により亜硝酸(NO_2^-)を二酸化窒素(NO_2)に酸化する蛋白質としてヘモグロビンとミオグロビンを同定した。また、凍結切片を用いて蛋白質チロシンのニトロ化を抗ニトロチロシン抗体で免疫染色を行うと、比較的低濃度(5〜10μM)のH_2O_2、NO_2^-で心筋細胞に局在が認められ、未知のペルオキシダーゼ活性を有した蛋白の存在が示唆された。そこで、蛋白変性剤を用いない二次元電気泳動後のニトロ化によるウエスタンブロット法を用いた新しい蛋白分離同定法を確立した。還元剤下では、ヘモグロビンはサブユニットに分かれ、その場合、チロシンニトロ化に関する高いペルオキシダーゼ活性を有していることが判明した。しかしながら、自然のヘモグロビン及びミオグロビンのチロシンニトロ化作用に必要なH_2O_2、NO_2^-はmM濃度であり、ヘモグロビンの分子変化の関与は不明である。一酸化窒素合成酵素(NOS)に依存しないNOの産生や非酵素的NO産生が、低酸素、pH5.5の条件で認められるので、ペルオキシダーゼを介したNO_2^-からNO_2の産生の関与を否定できない。現在、ラット虚血心又虚血再緩流後の心筋梗塞病変を用いて未知のペルオキシダーゼ活性を有する蛋白を検索中である。
