ヘリコバクタピロリ感染ネズミにおけるカビ毒素、食塩及び低V.C食による胃発癌実験

研究概要	これまでヘリコバクタピロリ感染とカビ毒素ステリグマトシスチンの相乗作用並びにVac A毒素の影響等について胃発がん作用について検討してきた。その結果、ステリグマトシスチンは胃粘膜にインテスティナルメタプレイシア,腸上皮化生即ち、前がん状態をヘリコバクタピロリ単独感染群に比し有意に発生することを明らかにしてきたが、明白な胃がんの発生は見られなかった。今回、更に、胃がん発生要因を明らかにするため、これまで疫学的に指摘されている高食塩及び農村癖地に多いヨー素欠乏のヘリコバクタピロリ感染との相乗作用としての胃発がんへの影響を明らかにするため、7週令のヘリコバクタピロリ感染スナネズミを用い、高食塩と低ヨー素並びに高食塩と低ヨー素同時組合せの群を作った。具体的には、CE-2標準食に5%食塩添加食22匹とCE-2から添加ヨー素を抜いた低ヨー素食群22匹及び低ヨー素,かつ5%高食塩の投与群40匹、ヘリコバクタピロリ感染のみの群16匹、ヘリコバクタピロリ非感染群も同様に5%食塩群21匹,低ヨー素群21匹,並びに5%食塩と低ヨー素群21匹,ヘリコバクタピロリ感染なしの群7匹の計170匹である。対照群21匹の各群間の胃発がん作用について比較検討する実験を進めている。 24ヶ月ないし30ヶ月の飼育期間中、各群の体重の測定を行い、成長曲線の作成、動物の行動や症状等の変化の観察を行っている。用いたヘリコバクタピロリの菌株はATCC43504である。スナネズミはセアック吉富(株)から購入。飼育室は準クリーンルームで室温は25℃、12時間毎の明暗リズムで飼育している。水と餌は自由摂取としている。餌は九動(株)から購入。餌の調整は3ヶ月毎としている。現在、ちょうど12ヶ月を経過したが食塩投与群の食事摂取量が多く、体重増加傾向がある他は特に外見上変化は見られていない。
