研究概要	p53やRBなどの癌抑制遺伝子の失活が発癌過程において本質的な役割を担っていることが明らかにされてきている。我々はp53-RB経路を再活性化させる薬剤を探索し癌の化学予防薬の開発を試みた。食物繊維の代謝産物である酪酸がp19遺伝子の発現を活性化することを報告していたが、その活性化機構をより詳細に検討した。p19の活性化にはプロモーターのSp1結合部位が関係し、転写抑制因子HDAC2がプロモーターから離れることが活性化の要因であると考えられた(Oncogene, Yokota, T.et al.)。また酪酸はp15の誘導も誘導することを明らかにし、この誘導もプロモーター領域を介しておこることを示した(FEBS Letters, Hitomi, T.et al.)。発癌プロモーターとして知られているTPAがp18遺伝子の発現を減弱させることを見い出した。この効果はpKC依存的であることをpKC阻害剤を用いて明らかにした。また、この発現抑制効果はp18遺伝子のプロモーター領域を介していることを明らかにした。これらの結果は発癌機構においてp18の量的減少が関与していることを示唆している(Oncogene, Matsuzaki, Y.et al.)。酪酸がp53下流遺伝子の一つDR5の発現を誘導することを見い出した。DR5のリガンドであるTRAILと酪酸の併用は相乗的に癌細胞にアポトーシスを誘導した。しかしながら、この併用は正常血球細胞に対してはほとんどアポトーシスを誘導せず、癌特異的に殺細胞効果を持つ可能性を示した(現在、投稿中)。食物フラボノイドの一つであるルテオリンがgadd45の発現を誘導することを見い出し、この効果はプロモーター領域を介していることを明らかにした。
