慢性関節リウマチ及びマウス関節炎モデルにおけるPD-1/PDリガンドの関与の研究

研究概要	我々が発見した抑制的共刺激分子PD-1/PDリガンド(PD-L)について、ヒト関節リウマチ(RA)でその異常が存在するか、PD-1/PD-Lシステムの異常がRA様の関節炎を誘導するかを明らかにすることを目的として研究を行っている。本年度の研究実績は以下の通りである。 1)RA患者関節液のCD4陽性T細胞にはPD-1陽性細胞が含まれていたが、末梢血やOA患者関節液には陽性細胞はほとんど無かった。このPD-1陽性T細胞はCTLA-4を発現しており、細胞内サイトカイン染色でIL-10を産生することから、何らかの抑制性機能を保有すると考えられた。また培養滑膜細胞をIFN-γなどで刺激してもB7など他の共刺激分子の発現は認めないのに比し、PD-Lは刺激に応じて強発現することを発見し報告した(Hatachi, et al.,2003)。 2)組織染色に適したモノクローナル抗体を多数作製し、ヒトRA患者関節組織でのPD-1やPD-L1発現を検討した。PD-L1は、RA患者関節の滑膜表層細胞や血管内皮に強発現していたが、関節浸潤細胞にもPD-L1発現細胞が多数存在していた。現在凍結標本を用いた二重染色法により、PD-1発現細胞のサブセットの同定や細胞内サイトカイン染色を行っている。 3)DBA1JマウスとB6マウスを用いて、タイプIIコラーゲン誘導関節炎(CIA)やコラーゲン抗体カクテルによる関節炎の発症実験を行い、後者では100%の発症率を得ている。CIAマウス関節組織においても、滑膜細胞はPD-L1陽性でありPD-L2は骨髄細胞の一部で陽性であった。CIAモデルマウスに抗PD-L1抗体を投与したところ、関節炎の発症が抑制される傾向を認めた。現在投与量や投与時期の検討、さらに抗PD-1抗体との同時投与実験などを準備している。 4)PD-1ノックアウトマウス(関節炎自然発症)を用いて、CIAの増悪/重症化を認めるかを調べる予定であるが、動物実験施設の感染症発生などのトラブルがあり、現在はSPF化などを行っているところである。SPF化後増殖させ、関節炎発症実験を行いPD-1/PD-L1の関節炎発症における制御的役割を解明する。
