胃上皮細胞の癌化過程における炎症性サイトカインと増殖因子の相互作用

研究概要

私共は、Helicobacter pylori感染を伴う皺襞肥大型胃炎において胃癌発生が高頻度であることに着目し、高ガストリン血症と胃粘膜内IL-1β産生の亢進が病態形成に果たす役割を明らかにしてきた。本研究において炎症粘膜から胃癌発生における炎症性サイトカインと増殖因子、ガストリンの役割を明らかにする目的で、1)ガストリン受容体を介するシグナル、2)胃癌細胞株およびH.pylori胃炎粘膜におけるDNAメチル化について解析した。
1)、ガストリンによるNF-κB化シグナルの解析
胃癌由来細胞株MKN28にヒトガストリン受容体cDNAを遺伝子導入した細胞株MKGRおよびモルモット単離壁細胞において、ガストリンはNF-κBを活性化してIL-8の産生を亢進させることを明らかにした。ガストリンによるNF-κBの活性化はPKC阻害薬やdominant negative PKC-δの導入により強く抑制され、PKC-δがガストリンによるNF-κB活性化に重要な役割を担っていることが明らかになった。NF-κBはIL-1シグナルにより活性化されることが知られており、ガストリンはIL-1と共通のシグナルを増強すると考えられる。サイトカイン刺激によるNF-κB活性化シグナルに関与することが明らかになっているIKK, NIK, TRAF2およびTRAF6のdominant negative変異体をMKGR細胞に導入して、ガストリン刺激によるNF-κBの転写活性に与える影響を検討した結果、ガストリンによるNF-κBの活性化にはTRAF6, NIKおよびIKKが関与しており、PKC-δはTRAF6/NIK/IKKカスケードの上流に位置することが明らかになった。
2)胃癌細胞株およびH.pylori胃炎粘膜におけるDNAメチル化の検討
胃発癌過程におけるDNAメチル化の意義を明らかにするため、胃癌細胞株およびH.pylori胃炎粘膜におけるDNAメチル化について解析した。胃癌細胞株においてTHBS-1やDAPkinaseなどの遺伝子のメチル化を高頻度に認め、H.pylori胃炎患者の生検材料を用いた検討において胃粘膜にDNAメチル化を高頻度に認めることが明らかになった。
