消化器癌治療への応用をめざした生存シグナル活性化機構の解明

研究概要	細胞死は多くの場合、FasやTNFαなどからの死のシグナルがcaspase cascadeを活性化することによりもたらされる。このcaspase cascadeの活性化を調節するものがインスリンやIGF-1などからの生存シグナルである。生存シグナル伝達分子であるAktやAkinaseは、Bc1-2ファミリーの1つでアポトーシス促進的に働くBADを燐酸化し、アポトーシス抑制的なBc1-XLとの複合体より解離させる。その結果、Bc1-XLがミトコンドリアからのチトクロームCの遊出を阻害してcaspase cascadeの活性化を抑える。ヒト原発性肝癌(以下、肝癌)組織20症例における解析では、AktやAkinaseは非癌部(NT=慢性肝炎or肝硬変)に比し癌部(T)で活性が有意に上昇しており、BADの燐酸化もTで増強していた。またアポトーシス最終実行分子であるcaspase3活性は80%の症例でTにおいてNTに比べて低下しており、その活性とBAD燐酸化との間には負の相関関係が認められた。これらの結果より、ヒト肝癌組織では生存シグナルがアポトーシス抵抗性に関与している可能性が示された。一方、MAPK superfamilyのヒト肝癌組織における活性化を検討すると、生存シグナルに反して癌部(T)で非癌部(NT)に比して有意にその活性が低下していた。そこでヒト肝癌細胞株において生存シグナル、p38MAPKの役割を検討した。10%FCS存在下では24時間後の細胞数は約1.3倍に増加したが、LY294002 (PI-3 kinase阻害剤)あるいはH89 (A kinase阻害剤)の添加で生存シグナルを抑制すると、細胞数は約70%に減少した。またLY294002やH89の添加により、BAD燐酸化は減弱しcaspase 3活性は逆に上昇した。細胞周期の解析と合わせると、生存シグナルであるAktやA kinaseはBADの燐酸化を介してcaspase 3活性を抑制し、肝癌細胞のアポトーシス抵抗性に関与する事が明らかになった。さらに生存シグナル阻害剤との抗癌剤との併用を行うと、抗癌剤の殺細胞効果が増強した。一方、p38MAPKの阻害剤、あるいはp38MAPKを活性化するMKK6の恒常的活性体を用いて、p38MAPK活性化が肝癌細胞株をアポトーシスに誘導することを見出した。以上の成績より、生存シグナルの活性化とp38MAPKの活性低下が肝癌細胞におけるアポトーシス抵抗性の一翼を担っており、その機能制御は癌治療の新たな治療法の開発に結びつくものと考えられた。
