消化管上皮幹細胞の同定と幹細胞を用いた炎症性腸疾患に対する粘膜再生療法の開発

研究概要	腸管上皮幹細胞は全ての腸管上皮細胞に分化する多分化能を有しており、また、体内の中でも非常に増殖能の高い細胞の一つである。精力的な研究が行われてきたにもかかわらず、その存在部位が腸管陰窩であることを除き、純化、機能解析は困難であった。近年、造血細胞や筋細胞においてDNA結合色素hoechst33342の排出能を特徴とするSP(side population)細胞に幹細胞が濃縮されることが報告され、普遍的な組織幹細胞純化法として注目されている。今回、我々はこのhoechst3332染色を用いることにより、腸管上皮幹細胞の純化を試みた。マウス腸管より採取した腸管上皮細胞よりSP細胞を解析したところ、腸管上皮においてSP細胞の存在が確認された。腸管上皮幹細胞の多く存在する陰窩上皮ではSP細胞が高率に認められたことに対し、絨毛上皮ではSP細胞は認められなかった。ABCG2の発現はSP細胞とMP(main population)細胞の間に有意な差を認めなかった。また、c-kit、Sca-1は腸管上皮細胞では発現が認められなかった。腸管上皮幹細胞の増殖・分化に重要とされるβ-catenin、神経幹細胞マーカーであるmusashi-1の発現はSP細胞において増加していることを見出した。これらの結果により、SP分画を用いることで、これまで困難とされてきた腸管上皮幹細胞の純化の可能性が示唆された。また、腸管上皮幹細胞において、Wntシグナルの下流にあるβ-catenin、Notchシグナルの制御因子であるmusashi-1の発現が増強していることより、これらのシグナルの協調作用が腸管上皮幹細胞の増殖、分化にとって重要であることが示唆された。今後、我々はこの方法を用いることにより、腸管上皮SP細胞のin vivo、in viroにおける機能的な解析を施行し、これらの細胞による粘膜障害に対する粘膜再生治療を検討したい。
