Wiskott-Aldrich症候群責任遺伝子WASPと会合する新規分子の解析

研究概要	我々は、Wiskott-Aldrich症候群患者50人を、その原因遺伝子WASPを解析することにより遺伝子診断し、臨床像を検討した。その結果、血小板機能低下による出血症状が予後不良因子である事を示した。そこで、WASP異常により血小板の機能低下が起きる機構を検討した。WASPがCIB(calcium integrin binding protein)と結合することを見出した。CIBは接着因子αIIbβ3インテグリンと結合する分子である。αIIbβ3は血小板に特異的に発現し、血小板凝集において中心的な役割を果たしている。血小板をトロンビン刺激すると、WASPとCIBは結合してWASP-CIB複合体を形成することが、免疫沈降による解析で明らかになった。そこで、CIBのN末端部分のペプチドを合成して血小板に導入し、WASP-CIB複合体形成を阻害したところ、血小板はαIIbβ3のリガンドであるFibrinogenとの結合が低下した。また活性化αIIbβ3を認識する抗体であるPAC-1による染色が低下した。すなわち、WASP-CIB複合体形成がαIIbβ3の活性化に重要な役割を果たしていると考えられた。次に、WAS/XLT患者で見られる変異を持ったWASPタンパクを作製し、CIBとの結合を検討したところ、変異WASPはCIBとの結合が低下していた。また、WAS/XLT患者血小板のトロンビン刺激後のPAC-1発現を検討したところ、著明に低下していた。すなわち、変異WASPはCIBと結合できず、WASP-CIB複合体が形成されず、その結果αIIbβ3が活性化できない事が明らかになった。以上の結果から、Wiskott-Aldrich症候群患者で見られる血小板機能低下は,接着因子αIIbβ3活性化障害が関与していると考えられた。
