デスモソーム異常に起因する自己免疫性および遺伝性皮膚疾患の病因解明

研究概要	プラコフィリン1に関しては、skin fragility-ectodermal dysplasia syndromeにおいてプフコフィリン1の遺伝子異常を見いだすことが証明されている。今回は、さらに多くのskin fragility-ectodermal dysplasia syndromeの家系を検索し、その遺伝子変異を同定し、Genotypeとphenotypeに関連があることを見いだした。デスモコリンに関してはIgA天庖瘡においてSPD型の抗原がデスモコリンであることを見いだした。さらに、免疫電顕を用いた検討から、SPD型の血清がデスモソーム部に反応するのに対して、IEN型は、非デスモソーム部に反応することを見いだし、IEN型の自己抗原がデスモソーム蛋白ではない可能性を証明した。エンボプラキン・ペリプラキンに関してはエンボプラキンおよびペリプラキンにcDNAを有核細胞発現系ベクターに組み込み、これらを培養表皮ケラチノサイトにトランスフェクトし、その動態を共焦点レーザー顕微鏡にて検討することにより両蛋白のケラチン中間径線維と結合することを見いだした。さらに、エンボプラキンおよびペリプラキンのリコンビナント蛋白を用いて腫瘍随伴性天庖瘡患者血清の反応を観察し、患者血中の抗エンボプラキンおよびペリプラキン自己抗体の反応を確認した。デスモヨーキンに関しては、すでに作成したデスモヨーキンノックアウトマウスを用いて、その形態学的観察および細胞接着能の検討からデスモヨーキンの細胞接着における重要性を検討した。その結果、デスモヨーキンノックアウトマウスは特別なphenotypeを示さず、光顕レベル、電顕レベルで、デスモソームの異常を見いださなかった。この事実は、デスモヨーキンのみの欠損ではデスモソームの大きな変化を引き起こさないことを示すものであった。
