造血細胞の生存と増殖・分化の制御機構に関する研究

研究概要	造血因子受容体下流のJAK/STATs系は、造血細胞の生存と増殖・分化に重要な役割を担っている。一方、多くの急性骨髄性白血病細胞において恒常的に活性化されているが、白血病原因遺伝子との関係については十分に解析されていない。急性前骨髄球性白血病(APL)では15、17転座によりPML/RARαキメラ蛋白が産生される。RARαはレチノイン酸により骨髄球系細胞の分化誘導、PMLはpremature senescenceやアポトーシス誘導に関与しており、転座によりPML/RARαが形成されるとRARαとPMLの両機能が阻害されAPLが発症すると考えられる。また、PMLは種々の転写因子と核内のnuclear bodyで複合体を形成し、それらの機能を制御することが報告された。一方、骨髄球系細胞におけるG-CSFを介した増殖、分化にはSTAT3が重要な役割を果たすことが知られている。そこで、PML/RARαならびにPMLのSTAT3に及ぼす影響について検討した。 Luciferase assayを用いた解析により、STAT3の転写能はPMLにより抑制され、逆にPML/RARαにより増強された。PMLはB-boxとCOOH-terminal領域を介してSTAT3と複合体を形成しSTAT3のDNA結合を阻害した。一方、PML/RARαはSTAT3と結合せず、STAT3からPMLを解離させることによりSTAT3の転写能を亢進させた。造血因子キメラ受容体(細胞外領域がG-CSF-R、細胞内がgp130)を発現させたIL-3依存性細胞株Ba/F3では、G-CSF刺激によりSTAT3依存性に細胞増殖が認められる。この細胞にPMLあるいはPML/RARαを導入し、G-CSF(STAT3)依存性増殖能に及ぼす効果について検討した。PML導入細胞ではG-CSF(STAT3)による細胞増殖が抑制されたのに対し、PML/RARα導入細胞では増殖反応が亢進した。以上の事から、PML/RARαによるSTAT3活性化亢進がAPL細胞の過剰増殖や異常分化に関与していると考えられた。
