ヒト慢性骨髄球性白血病の急性転化機構の解明と急性転化に関わる遺伝子の包括的解析

研究概要

慢性骨髄球性白血病(Chronic Myelogenous Leukemia : CML)は融合遺伝子産物p210BCR/ABLにより発症する造血器腫瘍であり、顆粒球の増多を主徴とする慢性期を経た後に幼若芽球の急激な増加を示す急性転化を起こす事を臨床的特徴とする。我々は造血前駆細胞に高発現している遺伝子のプロモーターを用いてp210BCR/ABLを発現するトランスジェニックマウスを作製し、世界で初めてのCMLトランスジェニックモデルの開発に成功した。更にこのマウスに癌抑制遺伝子p53のヘテロマウス(2対のp53遺伝子の一方を人為的に欠失させたマウス>を掛け合わせたところCML急性転化様の幼若芽球の増加を認め、我々の作製したトランスジェニックマウスはCML急性転化の解析においても有用であることを示された。このモデルマウスを用いてCML急性転化に関与する遺伝子を包括的に解析する目的で、トランスジェニックマウスにレトロウイルスを感染させ、内在性の遺伝子を活性化または不活化する「in vivo mutagenesis」を行った。実際には母乳を通じて産仔にウイルスが伝搬されるBHX2マウスをCMLトランスジェニックマウスと掛け合わせ、ダブルトランスジェニックマウスを作製した。マウスのgenetic backgroundをBHX2に近づける必要があり、そのため3世代のBHX2へのbackcrossを行った。現在BHX2へのbackcrossが世代終了したマウスが生まれており、末梢血の血球数および血液像を定期的に観察して結果、これまで5匹のp210BCR/ABL陽性マウスに急性白血病の発症を確認した。これらのマウスの腫瘍組織からDNAを抽出し、レトロウイルス特異的なプライマーを用いてPCRを行い、複数の急性転化発症の候補遺伝子を単離した。
