成長ホルモンの功罪に関する機序の解明:シグナル下流関連遺伝子の同定の解析

研究概要

成長ホルモン(以下GH)の抗老化作用について検討するため、各種GH標的細胞を用いて、GH刺激後誘導されてくる遺伝子または抑制される遺伝子を包括的かつ網羅的に探策した。GH刺激により発現が変動することが確認されたものについては、in vitro,in vivoで機能解析をした.
GHの標的臓器である血管平滑筋細胞、脂肪細胞、肝細胞、筋細胞のモデルを用いてGH刺激し、DNAマイクロアレイおよびDifferential Display法を使用し、GH刺激前後で発現が変化する遺伝子群をスクリーニングしたところ、血管平滑筋細胞ではミトコンドリアDNAがコードしているチトクローム酸化酵素および未知の数個の遺伝子が単離されてきた。このミトコンドリア遺伝子発現の増加は、ミトコンドリア遺伝子の転写および複製を制御する転写因子であるmtTF-1遺伝子発現がGHによって促進された結果であることを見出した。さらに、このmtTF-1の発現促進にはGHの細胞内シグナルに重要なJAK2を介するシグナルカスケードが関与することを明らかにした。この結果は、GHの抗老化作用と、老化と密接に関わっているミトコンドリア遺伝子の発現調節機構との間に、GH作用特異的なシグナルからミトコンドリア遺伝子発現を介した経路が関与するという新たな視点を提供した点で意義深い。現在mtTF-1遺伝子発現に対するGHの調節機構についてさらに詳しく検討を進めている。
脂肪細胞では脂肪酸合成酵素および未知の十数個の遺伝子が単離された。これらの遺伝子の中には、脂肪細胞の分化とともにその発現が変化する未知の遺伝子がいくつか含まれていた。得られた未知の遺伝子の機能解析についても順次進めている。
現在、既知・未知を問わず重要性が推測される分子については、ノックアウトマウスの作製に者手し、その分子の生理的機能の解析をめざしている。
