研究概要

PAX4/PAX6システムの人為的制御による膵β細胞分化誘導の試みの第一段楷として、本年度は、膵発生の過程において転写抑制因子として機能するPAX4に結合すると考えられるcofactorをyeast-two-hybrid法にて同定することを試みた。
まずYeast-two-hybrid法でのLibrary screeningに使用するGAL4 DBD(DNA binding domain)との融合蛋白質(bait plasmid)の構築を行った。Pax4の転写調節領域をbaitとして用いるが、この領域にはN末側に転写活性化領域とそれに隣接するC末側に転写抑制領域が存在する。このためbait plasmidにはそれ自身に転写活性化能が現れる可能性がある。そこで、Pax4転写調節領域の各種deletion productとGAL4 DBDとの融合蛋白質(PAX4/DBD-bait plasmid)を構築した後、これらの中で、転写抑制機能が保存され、かつ転写活性化能が認められなかったbait plasmidを選択した。
続いて、PAX4の転写抑制機能が著明に認められた細胞株(HeLa)から作成されたcDNA libraryとこのbait plasmidをmatingさせることで、library screeningを行った。その結果、約100個のpositive cloneが得られた。そして、これらのcloneからlibrary plasmidのみの回収を行い、それらを再度bait plasmidとyeast株でinteractionさせることで、非特異的結合をしていたlibrary plasmidを除外し、20個のpositive library plasmidを得た。
現在、このlibrary plasmidをシークエンスし、既知の遺伝子とのhomology検索を行いcandidate geneの絞込みを行っている。
