大腸癌で高頻度に認められる第18染色体短腕末端付近の特定アレル欠失の意義の解明

研究概要	正常大腸粘膜13サンプルと大腸癌組織140サンプルから得たcDNAについて、TS遺伝子の非翻訳領域に存在する繰り返し配列を挟んで増幅を行うプライマーT7-TS2を用い、T7 RNA polymeraseの存在下でPCRを行なった。153サンプル中正常大腸粘膜のTS遺伝子型が2R/3Rのヘテロであった54サンプル(35%)のうち33サンプルに、サテライトマーカーD18S59を用いたLOH analysisにより、TS遺伝子座を含むアレルの欠失が確認された。内訳は。2R/lossが13サンプル(24%)、3R/lossが20サンプル(37%)であった。正常大腸粘膜が3R/3Rもしくは2R/2Rのホモである大腸癌組織についても、TS遺伝子座を含むLOHの存在を観察した。その結果、正常大腸粘膜が2R/2Rである大腸癌組織では、5/9(56%)に、3R/3Rである大腸癌組織では、54/90(60%)にTS遺伝子座を含むアレルの欠失が認められた。したがって、TS遺伝子型はLOHの頻度に影響を及ぼさないものと考えられた。これらより、正常大腸粘膜のTS遺伝子型に関わらず、大腸癌組織で60%にTS遺伝子座を含む18pの欠失が存在することが明らかになった。次いで、D18S59以外に5種類の18p11マーカーD18S476、D18S481、D18S52、D18S452、D18S453と18p12.3のマーカーD18S57を使用し、18pの欠失範囲の判定を行なった。その結果、92サンプルに認められた18pの欠失はいずれも広範囲に及び、その領域にコードされている蛋白の発現は、LOHの有無と相関しなかった。当該領域に新規癌抑制遺伝子が存在する可能性は低いものと考えられる。
