精子形成関連遺伝子の単離とその解析

研究概要	全ての多細胞生物個体は体細胞と生殖細胞より成り立つが、前者は個体維持のために、後者は種の保存に重要である。この2種類の細胞間に存在する異なる法則性、特に生殖細胞特異的な法則を見つけその生物学的意義を明らかにする事は重要である。我々は哺乳類の生殖細胞なかでも減数分裂後に出現する半数体精子細胞の特異的な形態と特殊化した機能(運動能)を持つ精子に分化する過程の解析、即ち雄性生殖細胞にのみ存在する特異的な生命現象に研究の焦点を絞っている。このメカニズムを理解するには、半数体精子細胞特異的に発現する全遺伝子をクローニングし、そのコードする蛋白質の機能を解析する事が重要である。この為、成熟マウス精巣cDNAから幼若(半数体精子細胞出現前の生後17日目)マウス精巣cDNAを差し引いたライブラリーより、半数体精子細胞に特異的な全cDNAをクローニングし、各遺伝子のコードする産物の性質、組織及び細胞内での特異的発現を明らかにした。またgenomeDNAを単離し特異的発現の制御機構を調べる。更に、それらの遺伝子の欠損マウス、過剰発現マウス等を作成し、それらを統合して単離した遺伝子の半数体精子細胞分化にはたす役割を解明した。さらに、これらの遺伝子のヒト相同遺伝子を単離し、その産生する蛋白質の機能を解析するとともに男性不妊症とそれら遺伝子変異との関連を調べた。
