生体高分子に対する分子インプリントポリマーの新規調製法の開発と応用

研究概要

本年度は、昨年度に引き続き逆ミセル法による、水溶性化合物(生体高分子および低分子)に対する親水性粒子径単分散分子インプリントポリマーの調製を検討した。前年度に引き続きシラスポーラスガラスを疎水性化し、乳化媒体とした。また、本年度は、多孔質連続体膜として,水溶性高分子共存下,アルコキシシランの加水分解-重縮合反応によって作製した多孔質シリカ膜を疎水性化し、乳化媒体とした。まず、グリセリンジメタクリレートーアクリルアミド-メタノール-水溶液に、テンプレート分子としてオキシトシンを加えた溶液を分散相とした逆ミセル系エマルジョンを、シラスポーラスガラスを疎水性化した膜および多孔質シリカ膜を疎水性した膜を用いて膜乳化によって作製した。さらに、得られたエマルジョンのゲル化を、加熱処理によって行った。いずれの疎水性膜を用いても親水性均一性粒子の調製が可能であった。しかし、シリカ膜乳化のほうが,シラスポーラスガラス膜乳化より均一性の高い微粒子が得られた。一方、認識能をテンプレート分子を用いないで調製したノンインプリントポリマーと比較したところ、水系で、オキシトシンを認識できる親水性粒子径単分散分子インプリントポリマーの調製が可能であることがわかった。今後、より安定なポリマー微粒子調製の検討並びに粒子の量産化への検討を行うとともに、本法を生体高分子であるタンパク質あるいは核酸の分子認識に適用する予定である。
