Gタンパク質共役型受容体刺激応答の多様性を制御する分子メカニズムに関する研究

研究概要

Gタンパク質共役型受容体に作用するリガンド類は、様々な疾病の治療薬として応用されており、この受容体を介する情報伝達系を解明することは新たな薬物を開発していく上で重要である。本研究では、ヒトトロンボキサンA_2受容体(TP)と直接相互作用するタンパク質を同定するために、胎盤型(TP-α)および内皮型(TP-β)のcDNAのC末部分を作成し、酵母を用いたtwo hybridシステムによって、それぞれのC末部分と相互作用を示すタンパク質の検索を行った。その結果、プロテアソーム関連タンパク質であるproteasome subunit alpha type 3 (PSMA3あるいはα7)及びproteasome activator subunit 3 (PSME3、Ki antigenあるいはPA28γ)がTP-βと強く会合することを見い出した。一方、TP-αおよびTP-βにFLAGタグを付加したcDNAを作成し、それをCHO細胞に強制発現させたところ、FLAG抗体によって認識されるタンパク質の発現が見られるとともに、TP受容体刺激によるイノシトールリン脂質(PI)水解反応が惹起された。このPI水解反応はTP-α発現細胞では強く見られたが、TP-β発現細胞では極めて弱かった。以上のように、TP受容体にプロテアソーム関連タンパク質が直接会合していることを本研究によって初めて見い出し、TP-αおよびTP-βの機能的相違もこれら会合タンパク質が介在している可能性が考えられた。本研究の進展は、受容体制御機構におけるプロテアソーム関連タンパク質の新しい生理的意義を明らかにするものと期待される。
