ポリアミンとRNA、蛋白質との相互作用に基づく細胞機能のモジュレーション

研究概要	1.大腸菌においてポリアミンにより合成促進を受ける蛋白質として、オリゴペプチド輸送に関わるオリゴペプチド結合蛋白質、アデニル酸シクラーゼに次いで、RNAポリメラーゼσ^<38>因子を見出した。σ^<38>因子のポリアミンによる合成促進は、N末端から33番目の終止コドンUAGのread through促進によるものであった。このポリアミンによる促進は、UAG依存のsuppressor Gln-tRNA^<Gln>のリボソームへの結合がポリアミンにより促進を受けるためであった。 2.大腸菌では終結因子RF1のmRNAの26番目に終止コドンが存在し、+1 frameshiftによりRF2が合成される。このRF2合成に対するポリアミンの効果を検討したところ、ポリアミンは+1 frameshiftを促進しないことが明らかとなった。一方、マウスのアンチザイムmRNAの+1 frameshiftに対するポリアミンの効果を大腸菌で検討したところ、ポリアミンは真核細胞の場合と異なり強く+1 frameshiftを阻害することが明らかとなった。 3.ポリアミンはNMDA受容体活性を脱分極時に促進するが、その際のポリアミンの結合部位をsite-directed mutagenesisにより作製した変異NMDA受容体活性を測定することにより同定し、アゴニスト結合領域より更にN末端側にあるこの領域を調節領域(R-domain)と命名した。この約350アミノ酸残基から成るR-domain(His-tag付加)を大腸菌に発現させ、その性質を検討した。NR1Rはスペルミン及びイフェンプロジルと強く結合したが、NR2AR及びNR2BRはスペルミンとは強く結合したがイフェンプロジルとは全く結合しなかった。
