研究概要

Ca^<2+>の関与する様々な細胞機能に必須の役割を果たす細胞質内「モジュレータプロテアーゼ」カルパインは、その機能の重要さ故に活性不全により、筋ジストロフィー、虚血性疾患、がん、糖尿病、白内障、アルツハイマー病など様々な病態の発症に関与する。これらの病的状態において、カルパインの活性阻害が症状の軽減につながることが、様々な例で報告されている。カルパスタチンは内在性のカルパイン阻害タンパク質であり、その特異性、阻害活性において最も優れたものであるため、これを元にした改変タンパク質の利用が興味深い。本研究では、サブユニットへの解離と活性化の関係、その細胞内の部位の同定、そしてその活性化機構について、カルパスタチンとの関係にも注目しながら解析し、カルパインの活性化におけるサブユニットの解離に焦点を絞った解析を行い、カルパインの活性化の分子機構の解明を目指した。カルパインのプロテアーゼドメイン(II)を大腸菌に発現させて、その性質を調べ、また、培養細胞発現系などを用いてカルパインのサブユニット解離の動態を解析した。その結果、カルパインの自己消化は活性化に必要な過程ではなく、むしろサブユニットの解離を促進し、基質の特異性や局在などを変化させ、制御サブユニットに新たな生理機能をもたらす可能性を明らかにした。さらに、EGFPタグを付けたC105S不活性型m-カルパインなどを用いて、培養細胞とカルシウムイオノフォアをはじめとする様々な刺激により、カルパインの動態を解析した。その結果、フォドリンやタリンなどの細胞骨格系のタンパク質の切断に、m-及びμ-カルパインがディファレンシャルに関与しており、その切断は膜上で行われ、カルパスタチンではほとんど阻害されないことが明らかとなった。
