器官形成における細胞系譜制御機構の解明

研究概要	器官形成過程においては器官特異的幹細胞がその多能性を維持する機構、器官特異的幹細胞の分化の運命付けの機構、分化がコミットされた細胞系譜が他の系譜に向かわない運命付けの維持と分化の排他性の機構など重要な仕組みが存在する。本研究では神経系と造血系に的を絞って、これらの細胞系譜制御機構に取り組み、次の成果をあげた。(1)マイクロアレイ法を用いて、マウス中枢神経系前駆細胞の多分化能維持培養系と分化誘導培養系との間で発現が変化する遺伝子として同定した遺伝子群について順次マウス胎仔脳組織切片上での発現解析を実施し、神経幹細胞が存在する脳室下帯での発現を確認した。(2)昨年度までに見出したBMP2によるニューロン分化抑制シグナルと、従来報告されているニューロン分化抑制性のNotchシグナルとが、核内でクロストークして相乗的な遺伝子発現の誘導に至ることを新たに見出した。(3)神経上皮細胞培養系においてアストロサイト分化誘導性サイトカイン刺激後に発現が上昇する遺伝子群の解析をマイクロアレイ法で行い、CITED1と呼ばれる遺伝子を同定することができた。(4)胎生期マウス脳において、神経上皮細胞から胎生の進行に伴いアストロサイトが分化し始める時期に呼応して、アストロサイト特異的蛋白S100βの遺伝子プロモーターの特定のシトシン残基のメチル化が外れることを見出した。(5)胎生期マウス造血組織で発現する遺伝子として同定したELYS遺伝子欠損マウスの作成と解析により、この遺伝子が欠損することで胚盤胞期を超えて発生しないことがわかった。(6)胎生期マウスで成体型造血が始まるAGM領域の培養系を用いた実験から、Ras/MAPK経路を負に制御する分子であるSpred-2がAGM領域からの造血前駆細胞集団の拡大を負に制御していることがわかった。
