流動的な生体膜レセプターと多価相互作用する超分子リガンドの設計

研究概要	本研究では、多数のα-シクロデキストリン(α-CD)の空洞部を貴通したポリエチレングリコール(PEG)の両末端に嵩高い生分解性基を導入した串刺し状高分子(生分解性ポリロタキサン)を基本骨格として、多数のα-CD水酸基に種々のリガンドを導入し、α-CDのPEG鎖上での位置が自由に変化する超分子の特徴から、細胞表面レセプター発現・分布の時間変動に同調してレセプターとの結合を持続する新しい生体材料を設計することを目的としている。昨年度は,マルトース(Mal)導入量の異なるMal-ポリロタキサン結合体(Mal-PRX)を合成し、CD貫通数が約85(貴通率:38%)であるときにマルトースの分子運動性が高いことが見出すとともに、これら結合体のレクチン(コンカナバリンA : ConA)とMal-ポリロタキサン結合体による赤血球凝集阻害試験から、マルトースの百万倍にも相当することを見出した。この成果を受け本年度は、ConAとの結合定数を算出し、ポリロタキサンの機械的結合に基づく分子運動性が熱力学的にも有利に機能することを見出した。さらには、細胞表面での分子認識への応用を検討するため、リガンドにジペプチドであるバリルリジン(VK)を導入した。VKは小腸のペプチドトランスポーターPEPT1から選択的に細胞内へ輸送される基質であり、ポリロタキサンへの導入によってPEPT1からは吸収されずにペプチドやアミノ酸の輸送を阻害することが確認されている(Bioconj.Chem., 13, 582-587 (2002).)。上皮細胞上に発現するPEPT1の発現量が少ない時にはVK-ポリロタキサン結合体添加後の基質輸送を阻害しなかったが、発現量を多くすると著しい阻害効果が確認された。このことは、PEPT1がVK-ポリロタキサン結合体1分子と多価相互作用して基質の輸送を阻害したことを示唆する結果であり、従来の低分子阻害剤では見られない現象であった。これらのことから、リガンド-ポリロタキサン結合体1分子に多数のレセプターが結合するような分子形態と、ポリロタキサンの機械的結合に起因する分子運動性の確保が最も重要な因子であることが見出された。
