p53遺伝子欠損軟骨細胞を用いた軟骨分化制御機構の解明

研究概要	1.PGE2シグナルの解析関節軟骨におけるPGE2受容体の発現を免疫組織化学及びRT-PCRで解析した結果、EP2が主たる受容体であることが判明した。前年度までの研究により樹立したp53-/-マウス由来関節軟骨細胞であるMMA2をEP2アゴニストで処理すると、濃度依存性に細胞内cAMP濃度が上昇した。EP2アゴニスト処理前後の遺伝子変動を、マイクロアレイを用いて解析することで、EP2アゴニストシグナルの下流遺伝子群を同定した。その中には細胞増殖を促進し、アポトーシスを抑制する遺伝子が複数含まれていた。ヒト初代培養軟骨細胞にEP2アゴニストを作用させると、BrdU取込が上昇した。ラット大腿骨を用いた器官培養において、EP2アゴニストによる処理により、大腿骨骨頭関節軟骨組織中のPCNA陽性軟骨細胞数が増加した。以上よりPGE2はEP2を介して関節軟骨細胞の増殖を促進する作用をもち、EP2アゴニストは新規の退行性軟骨病態に対する治療薬として期待できることが判明した。 2.石灰化関連遺伝子の単離単層培養により石灰化結節を形成するMMR14と形成しないMMR17の間の遺伝子発現の相違をマイクロアレイを用いて解析した。その結果、ALPなど石灰化に直接関連すると考えられる遺伝子に加えて、いくつかの細胞外基質、細胞接着因子、シグナル伝達因子及び転写因子が同定された。その中で特に細胞接着因子であるOBカドヘリンに注目して解析した。MMR14では培養日数とともにOBカドヘリンの発現がmRNA及び蛋白レベルで上昇してくるのに対し、MMR17では全くそのような誘導が認められなかった。マウス肋軟骨における免疫染色ではOBカドヘリンは石灰化軟骨において陽性であり、最終的な石灰化に関与している因子の一つであることを示唆する結果が得られた。
