T細胞の分化と細胞死のシグナル伝達におけるレドックス調節機序の解明

研究概要	(1)酸化的化学修飾によるチロシンキナーゼ活性化の機序の解析:ヒトリンパ性白血病細胞から分離された受容体型チロシンキナーゼRETの分子モデルにおいて、がん遺伝子産物RET-PTC-1のキナーゼドメインのC末端側に保存された特定のシステイン(Cys376)をアラニン(疎水性)、グリシン(親水性)、セリン(親水性、OH 基保有)、リジン(塩基性、e-アミノ基保有)のそれぞれに置換したミュータント分子の遺伝子(cDNA)を導入した細胞を作製した。これらの細胞から分離したRETの活性をin vitro kinase assayで解析した結果、いずれのミュータントRET分子のキナーゼ活性も対照の親RET分子のそれに比べて極めて低値であり、この位置のシステインが酵素活性の発現に重要であることが確認された。そこで、RET蛋白のアミノ酸配列から北里大の梅山博士が開発した蛋白モデリングソフトによって3次構造を推定し、その中で、このシステインの反応基の役割を推測した。(2)T細胞表面への酸化ストレスにより起動するシグナル伝達の機序の解析:T細胞表面の受容体が砒素などで酸化的化学修飾を受けると、細胞の生と死を誘導する正反対の2種類のシグナルが起動すること、そのいずれも、細胞膜ラフトの機能に依存して産生が促進される活性酸素の働きを必要とすることを明らかにした。(3)T細胞の分化と細胞死のレドックス調節の機序におけるp38MAPK上流のMKK6の役割の解析:樹立したMKK6-/-マウスを解析し、胸腺リンパ球のネガティブセレクションとそれ以後の終末分化において、MKK6と関連するレドックスシグナルが関与することを明らかにした。
