IgG結合性をもつ超微量ヒト血漿タンパク質群の分離および構造解析に関する研究

研究概要	ヒト血漿タンパク質の非変性条件の2次元ゲル電気泳動(ゲル等電点電気泳動とサイズ分離ゲル電気泳動のいずれにも変性剤を用いない)によるパターンと、部分的変性条件の2次元ゲル電気泳動(非変性条件の等電点電気泳動とSDSゲル電気泳動を組み合わせる)によるパターンを比較することによって検出された微量タンパク質群について、その由来を調べた。また、これらのタンパク質群について、質量分析による構造解析を行った。質量分析による構造解析には、マトリックス支援レーザー脱離イオン化飛行時間型質量分析計MALDI-TOFMSによるペプチドマスフィンガープリント法と、エレクトロスプレーイオン化タンデム質量分析計(ESI-MS/MS)による部分アミノ酸配列決定法を用いた。さらに、平板型アガロースゲルを等電点電気泳動に用いる3段階電気泳動法を開発し、これが微量タンパク質を濃縮し質量分析で構造解析するための方法として有効であることを示した。これらの構造解析結果と、サイズ分離におけるタンパク質解離条件の異なる3種の2次元電気泳動分離パターンを比較することにより、ペプチドから機能を持つタンパク質が形成される過程を明らかにすることができると考えられる。
