研究概要

昨年までは,行動から細胞レベルまでを脳の階層性に沿って学習・記憶の機構を調べてきた.そこで本年度は分子生物学からのアプローチに的を絞った.
(1)軟体動物のモノアラガイを用いて,その中枢神経系内の単一ニューロンでのmRNAコピー数を測定する方法を開発した.モノアラガイに味覚嫌悪学習を施し,その学習の鍵を握るニューロンを単離して,長期記憶に関連する転写因子CREBのmRNA量を測定した.そして味覚嫌悪学習前後でそのmRNA量変化を調べたところ,抑制型のCREB2 mRNAが有意に減少していることが分かった.したがって,モノアラガイの味覚嫌悪学習の長期記憶は,CREB2の遺伝子発現の減少により形成されることが示唆された.
(2)CREB1は一般的に転写促進型と言われている.そこでモノアラガイのCREB1についてそのアイソフォームを調べてみた.その結果,CREB1には7つのアイソフォームがあることがわかった.しかし,そのうち4つは転写活性に必要なリン酸化部位を欠いていることが明らかとなり,CREB1アイソフォームも抑制型として働く可能性を示すことができた.
(3)CREBによって他の転写因子C/EBPが発現することがわかっている.そこでC/EBPの次の遺伝子を検索したところ,一酸化窒素合成酵素(NOS)が発現してくることがわかった.実際にモノアラガイの中枢神経系において,C/EBPが発現しているニューロンにはNOS活性があることを示すことができた.
