多数のチオストレプトン系大環状ペプチド抗生物質の初めての全合成と構造-活性相関

研究概要	最近、種々の菌類の二次代謝産物の中から大環状チオストレプトン系抗生物質やその類縁体が次々と発見され、単離・構造決定されている。それらはいずれも複素環や異常アミノ酸から構成された特異な構造をしている。また、そのほとんどが興味ある生理活性を示すことが知られている。我々はこの一連の抗生物質群の全合成および構造活性相関に興味をもち全合成を行い、この種の天然物としては先導的に初めてミクロコッシンPおよびP_1の全合成やGE2270Aの形式全合成に成功した。この1年間で上記の知見をもとにデヒドロペプチドからチアゾールアミノ酸やオキサゾールアミノ酸の重要な合成法を確立した。それらの合成方法を基に、未だ合成例のないThiocillineIの全合成に初めて成功した。さらにSulfomycin類の異常アミノ酸由来の重要フラグメントを合成しそれらのフラグメント縮合も行った。また、シクロチアゾマイシンについては最も合成が困難と考えられたα-S架橋を含むFragmentEおよびオキサゾリン-チアゾリン変換法を利用してFragmentDを合成し、FragmentA-E-Dの縮合にも成功した。次にSulfomycin類やベルニナマイシン類に含まれる2位にオキサゾールが置換した三置換ピリジン骨格の新規合成法を開発した。一方、最近、単離構造決定され抗癌剤として期待されているビストラタミド類も我々が新たに開発した方法で容易に全合成ができた。これらの知見は一連のチオストレプトン系抗生物質やその類縁体の全合成に役立つと確信している。
