腫瘍の放射線治療におけるアポトーシスの誘発に関する研究

研究概要

本研究は腫瘍の放射線治療時における腫瘍細胞のアポトーシス誘発感受性と種々の遺伝子異常との関連性を明らかにし、また、放射線治療時におけるアポトーシス誘発や正常組織障害の発生におけるサイトカインの役割を明らかにすることを目的として今年度は以下のような結果を得た。
1)腫瘍の放射線治療効果の解析:放射線治療効果に対するアポトーシス誘発の影響を解析するために、イヌ、ネコの個々の腫瘍例における治療効果について検討した。放射線単独または化学療法との併用時にリンパ腫、鼻腔腺腫、脳腫瘍、唾液腺癌、未分化肉腫などで良好な治療成績を得られた。一方、肺などに転移のある末期悪性腫瘍で、軟骨肉腫、悪性黒色腫、線維肉腫、扁平上皮癌などで治療効果が小さかった。
2)放射線治療時における副作用の検討:副作用におけるサイトカインの関与を検討するために、発生状況について検討した。線量が35Gyを越えると皮膚の発赤や炎症などの皮膚障害が認められる傾向にあり、犬で24%、猫で31%に脱毛、皮膚の色素沈着、皮膚の発赤や炎症などの急性の副作用を認めた。
3)アポトーシス誘発経路の解析:リンパ腫で良好な治療効果が得られたことから、ラット胸腺細胞やヒトリンパ腫株化細胞におけるアポトーシスの誘発の機序について検討し、同じリンパ腫でもアポトーシス誘発経路に差があることを示した。
4)サイトカイン検出・定量法の検討:炎症性サイトカインのIL-1_とIL-1_の生物活性を抑制する因子として重要な働きをするIL-1レセプターアンタゴニスト、可溶型IL-1レセプターのI型とII型を検出・定量するサンドイッチELISAを確立し、種々の炎症性疾患の病態を詳細に検討することが可能となった。
