糸球体足細胞の細胞膜ドメイン形成に関する細胞生物学的研究

研究概要

糸球体足細胞の足突起間にある特殊な細胞間接着構造であるスリット膜はapical膜とbasolateral膜を境界する構造として、膜ドメイン形成に重要な役割を演じていると考えられるが、それに関わる分子群は明らかではない。本年度は、新たなスリット膜関連蛋白としてポドシンを新潟大腎研のグループとともに同定した。また、線虫で見出された極性蛋白par3の哺乳類ホモログであるASIPが、尿細管上皮細胞ではタイト結合の頂部側末端に位置し、apical膜の境を規定していることを見出した。さらに、このASIPが糸球体足細胞のスリット膜基部に発現しており、スリット膜を境とする膜ドメイン形成に中心的な役割を演じていることを明らかにした。また、塩野義研で見出された新規先天性ネフローゼラットの病態解析を行い、このラットがネフローゼを発症する初期に、スリット膜関連蛋白であるZO-1を安定して発現しているが、ネフリン、ポドシンおよびASIPの発現が著しく低下することを見出した。次に、温度感受性変異を入れたSV40T抗原遺伝子のトランスジェニックラットより単離した足細胞培養株の特徴づけを行い、得られた細胞が33度で突起形成能を有し、突起を近隣の細胞に延ばした状態で分裂をすること、その際、突起の結合部にはASIPが発現していることを見出した。また、37度で細胞増殖を止めた際に認められる足細胞特異的なマーカー蛋白の発現の変化を各クローンで詳細に検討し、足細胞の足突起に発現するアクチン結合蛋白シナプトポディンの発現が有意に増加し、またアクチン繊維とアクチン結合蛋白α-アクチニンの再配列が起こることを確認した。これは得られた多くのクローンで共通した性質であり、足細胞培養系での温度変化に伴う良い指標となると考えられる。
