ゲノム情報を用いたヘテロ二量体型アミノ酸トランスポーターファミリーの全容の解明

研究概要

本研究は、研究代表者らが分子実体解明を推進してきたヘテロ二量体型アミノ酸トランスポーターファミリーのゲノム上の全メンバーを明らかにし、その機能を同定することにより、ヘテロ二量体型アミノ酸トランスポーターファミリーの全容を明らかにすることを目的として行われている。ヘテロ二量体型アミノ酸トランスポーターファミリーは、トランスポーターの本体である12回膜貫通型のタンパク質(主サブユニット)と、1回膜貫通型の補助因子(補助サブユニット)が、ジスルフィド結合によって連結することにより形成される。主サブユニットはヘテロ二量体型アミノ酸トランスポーターファミリーのSLC(solute carrier)7ファミリーに属する。平成14年度は、既知のSLC7ファミリーのcDNA配列を用い、公開されたヒトゲノム及びマウスゲノム塩基配列を検索し、このファミリーに属するさらに4種の未同定の遺伝子(暫定的にSLC7-N1〜4と命名)が存在することが明らかにした。4種のcDNAの全長cDNAクローンを得て、DNAシーケンシングを行ったところ、SLC7-N1及びSLC7-N2は、14回膜貫通構造を持ち、SLC7ファミリーの既知の塩基性アミノ酸トランスポーター群に類示すること、SLC7-N3は12回膜貫通構造を持ち、未知の補助サブユニットと連結するAsc-2と比較的高い相同性を有すること、SLC7-N4は、6回膜貫通構造を持ち、ヘテロ二量体型アミノ酸トランスポーターファミリーの他のメンバーの半分のサイズの膜タンパク質であることが明らかになった。アフリカツメガエル卵母細胞に発現させることにより、SLC7-N1は、アラニンをNa^+の存在化で輸送する小型中性アミノ酸のNa^+依存性トランスポーターであること、及びSLC7-N2は、アルギニンを中心とする塩基性アミノ酸のNa+非依存性トランスポターであることが示された。
