糖鎖欠損マウスを用いた神経変性・再生のメカニズムと責任遺伝子の解析

研究概要	GM2/GD2合成酵素遺伝子欠損(KO)マウスでは、病理所見として加齢に伴う坐骨神経、脊髄の神経系細胞の変性が認められる。GM2/GD2合成酵素遺伝子KOマウスの運動及び知覚機能を検討した結果、1)Rota-rod法による協調運動異常、2)加齢に伴う歩行異常(Foot print法)、3)機械刺激に対する感受性の低下などが認められた。そこで、複合型ガングリオシド欠損マウスにおける神経線維の異常の発症機構を分子レベルで解明する為に、まず、GM2/GD2合成酵素遺伝子KOマウスについて、野生型マウスと発現レベルの異なる遺伝子をcDNA subtraction法及びDNAチップにより検討した。(1)subtraction法:現在までに、GM2/GD2合成酵素遺伝子KOマウス及び野生型マウス,(20週齢、雄)の脊髄mRNAを用いて、subtractionを行い、KOマウスにおいて発現が消失する遺伝子をクローン化した。これらのクローンをdot blotしたメンブレンアレイを作製し、KOマウス及び野生型マウスの脊髄mRNAからcDNAプローブを作製し、hybridizationを行った結果、約300クローン中2クローンがKOマウスで特異的に減少してた。現在これらの遺伝子の同定とNorthern blottingよる発現レベルの変化を確認中である。(2)マイクロ・アレイ:マウスoligo DNAチップ(約1万遺伝子)を用いてKOマウスと野生型マウスの間で発現レベルの異なる遺伝子を検索した結果、ある種のサイトカインにより誘導される数種の遺伝子発現増強がKOマウスにおいて認められた。また、KOマウスにおいて3〜5種の遺伝子で発現低下が認められた。現在、RT-PCR及びNorthern bottingよる発現レベルの変化を確認中である。
