糖鎖欠損マウスを用いた神経変性・再生のメカニズムと責任遺伝子の解析

研究概要	高齢化社会の到来に伴い、種々の神経変性疾患の原因究明とその治療法の開発が課題となっている。我々は、神経系に多く発現している酸性スフィンゴ糖脂質の糖転移酵素遺伝子のクローニングを行い、神経系におけるガングリオシドの機能を解明するために種々の糖転移酵素遺伝子の欠損マウス(KOマウス)を作製し、糖鎖欠損マウスの異常に関する解析を行ってきた。その結果、複合型ガングリオシドは神経系組織の維持と生存及び神経修復にとって必須の分子群であると考えられた。本研究では、GM2/GD2合成酵素遺伝子KOマウスについて糖鎖変異マウスの加齢に伴う異常の検討をふまえて、神経系細胞の生存及び損傷修復に重要なガングリオシドの作用メカニズムをマイクロアレイなどの手法を用いて包括的に分子レベルで解析した。DNAマイクロアレイにより20週齢オスの野生型とKOマウス脊髄における遺伝子発現の差異を検討した結果、TGTP、IFI47、IFIT1、IRF7及びIRF1においてKOマウスで発現の上昇傾向が認められたが有意の差はなかった。今回の実験で有意の発現変化を示す遺伝子が同定できなかった原因として、本KOマウスのように長期にわたり緩慢に神経変性が進行する場合においては、劇的に変化する遺伝子を同定することが困難である可能性が考えられる。また、脊髄後角においては明らかな神経変性が認められるが、実験試料として全脊髄を用いたことが検出感度を低下させ、変性関連遺伝子が検出できなかった一因と考えられる。
