ヒトNKT細胞前駆細胞の解析と体外増殖法の開発

研究概要

CTL, NK, NKT細胞やDC等の免疫担当細胞の役割を総合的に解析し、がんの治療法の確立に貢献することを本研究の目的とする。NKT療法の臨床応用を目指し、臨床での使用が可能なヒトNKT培養法の確立ならびにその評価法の確立を行った。我々はG-CSF投与後の末梢血或いはアフェレーシス血から得られた単核球を用いることによって、通常の単核球より効率よくVα24^+NKT細胞が誘導可能であることを示してきた。これらの培養においては研究用のRPMI培地やFBSを用いていた。臨床応用を考える場合、FBSは好ましくないので、ヒト血清・血漿並びにヒトアルブミンや臨床用細胞培地(AIM培地)を用いた培養を試みた。
その結果、1.AIM培地でもRPMI培地と比較して遜色なく培養が可能であることとともに、ヒト血清・血漿を用いれば、FBSを用いる以上に効率よくVα24^+NKT細胞の誘導が可能であることが明らかとなった。
2.G-CSF投与後の細胞を用いることで、Vα24^+NKT細胞の増殖は有為に増強された。
3.G-CSF投与後の血漿を用いることで、Vα24^+NKT細胞の増殖は有為に増強された。
4.G-CSF投与後の細胞と血漿の組み合わせは、Vα24^+NKT細胞を最も効率よく増殖させた。
5.G-CSFの投与前の血漿と比較して、G-CSF投与後の血漿中には、G-CSFと共にIL-3とIL-13が高濃度に含まれていた。
6.培養ヒトVα24^+NKT細胞を含む細胞群は、各種ヒト培養がん細胞に対して高い抗腫瘍活性を示した。この効果は、標的細胞のCD1dの発現量に左右されなかった。
7.6.の細胞群から、MACSを用いて95%以上に純化したVα24^+NKT細胞も、各種がん細胞株に対して高い抗腫瘍活性を示した。この効果も、標的細胞のCD1dの発現量に左右されなかった。
8.6.の細胞群から、MACSを用いて95%以上に純化したVα24^+NKT細胞は、IFN-γを高濃度に産生した。
