大腸癌組織浸潤リンパ球が認識する抗原群のクローニングと解析

研究概要

【日的】ヒトの免疫系、特に腫瘍浸潤リンパ球(TIL)由来の細胞傷害性T細胞(CTL)株が認識する分子群が機能分類上どの様な分子種を認識しているのかを探索するために本研究を行った。
【実験成績および考察】大腸癌TILから樹立したOK-CTL株が認識する遺伝子群125種類を単離・解析した。
(a)塩基配列を決定し、相同性の検索、点突然変異の有無を解析した。さらに、タンパク質2次構造、モチーフ等はNCBIのデータベース解析を行い、機能を推測して遺伝子の分類を行った。遺伝子の種類が多いことから、データベースを用いたバーチャルノーザン(SAGE)解析によりおこない、未知遺伝子の発現はノーザンブロットおよびRT-PCRで行った。その結果、重複13種類、断片遺伝子40種類、cDNA濃度依存性CTL認識が確認できなかったもの26種類で、既知タンパク質もしくはその断片、あるいは機能モチーフを有する未知遺伝子46種類を選別した。これらの遺伝子のうち、細胞増殖に関与する、または増殖の結果発現亢進する自己抗原をコードする遺伝子、および分子の運搬に関与する分子が多いことがわかった。(b)上記遺伝伝子がコードするタンパク質のアミノ酸配列から、HLA-A2結合モチーフを持った9-10残基のオリゴペプチド配列を526種類委託合成し、HLA-A2をもったT2細抱株にペプチドを感作して標的細胞としてOK-CTLがペプチドの濃度依存性に認識した208種類のペプチドを選別した。(c)癌浸潤リンパ球が特異的に認識する事が証明されたペプチドをコードする遺伝子の機能を、各種データベース検索による推定し、免疫系が生体で認識している自己抗原の機能別分類を行った。
【結語】TIL由来のCRL株が認識する分子群は、細胞増殖に関与、または増殖の結果発現が亢進する自己抗原遺伝子が多く、次に分子の運搬に関与する分子が多いことがわかった。
