遺伝子導入による血管内皮細胞機能制御と虚血性疾患の遺伝子治療への展開

研究概要	血管内皮細胞の機能、増殖を制御することで血管病の遺伝子治療を目指し、血管新生に関わる生理活性物質のVascular endothelial growth factor(VEGF),Angiopoietin 1(Ang 1),Prostacyclin合成酵素、各遺伝子をAAVベクターへくみこみマウスの骨格筋に投与し血管新生の有無を検討した。AAVベクターでは血清型により臓器、細胞での遺伝子導入発現効率がことなるため、発現効率を各血清型AAVでマーカー遺伝子や凝固第IX因子遺伝子などをもちいて検討すると筋肉で導入遺伝子発現効率はAAV1がもっとも良いことがわかった。そのため、AAV1ベクターへこれらの遺伝子を組み込みマウスの下肢筋肉に投与し、血管新生を比較した。VEGFのみを発現させたマウスでは筋繊維間の血管の増加が認められたが、VEGFとAng1を発現させたマウスでは正常にてみとめられる筋繊維間の毛細血管以外に、束状に特殊な新生血管が形成された。この新生血管は血管内皮細胞のマーカーであるフォンビルブランド因子、VE-Cadherinが陽性の血管内皮細胞の周囲を、壁細胞と思われる細胞がかこんでおり、強固な血管新生と考えられた。また、この新生血管は筋肉内のみならず筋肉外組織にも認められ、新たな血管形成の可能性が示された。VEGF、Ang1に加え、Prostacylin合成酵素を発現させると血管新生はさらに増強しており、種々の血管新生に関わる生理活性因子を発現させることで虚血に対しての遺伝子治療のあらたな可能性が示唆された。
