遺伝子導入による血管内皮細胞機能制御と虚血性疾患の遺伝子治療への展開

研究概要	研究計画に基づいて、血管内皮細胞特異的に発現させるためにPAI-1プロモーターの下流にマーカー遺伝子、血管内皮増殖因子遺伝子、抗血栓性因子遺伝子などを搭載したウイルスベクターを作成し実験をおこなった。in vitroで分化誘導し、血管内皮細胞のマーカーを発現するES細胞にレトロウイルスベクターをもちいてGreen Fluorescent Protein (GFP)遺伝子と血管内皮増殖因子を導入した。遺伝子導入したES細胞をSCIDマウスへ移植し生体内においてES細胞由来の細胞による血管新生がおこるかを検討したところ、一部にGFP陽性で、内部に赤血球などの血球細胞を認める管腔構造を確認できた。PAI-1プロモーター、あるいはCytomegalovirus (CMV)プロモーターの下流にLac Z遺伝子、凝固因子遺伝子を搭載したType 1アデノ随伴ウイルス(AAV1)ウイルスベクターを作成し、マウスへ骨格筋に投与し遺伝子発現を検討したところ、CMVプロモーターを用いたときには、骨格筋内に遺伝子産物が検出されたが、PAI-1プロモーターにより遺伝子を発現したときには、骨格筋ではなく、血管内皮細胞のマーカーの一つであるPECAM-1の発現と一致する導入遺伝子の発現が骨格筋周囲にあり、血管内皮細胞に特異的に遺伝子導入を行えたことが確認できた。さらに、マウス血液中にヒト凝固因子遺伝子を導入したときは、ヒト凝固因子がマウス血液中に検出された。さらに、抗血栓性因子遺伝子を搭載したベクターを投与したときには虚血を軽減することが示された。以上の結果から、ウイルスベクターを用いて機能遺伝子を導入することで内皮細胞機能を制御し血管新生、血管病変治療を行いうる可能性が示唆された。
