皮膚樹状細胞における皮膚プリオン蛋白発現の制御機序の解析

研究概要

プリオン病は、プリオン蛋白(PrPc)が変換してプリオン変異蛋白(PrPsc)を誘導することによって発生する疾患であるが、本疾患の原因の一つとして、皮膚内にPrPを投与することによって、PrP病を誘導することができる可能性が明かにされている。皮膚においてLCは、種々の抗原を所属リンパ節に誘導することができる抗原提示細胞であり、PrPの伝播に関与する可能性が示唆される。これまですでにマウス表皮から高純度でLCを採取することに成功している。高純度のLCを用いた検討では、精製LCがPrPcを発現することを明らかにした。次にサイトカインによってLCのPrP発現の変化を受けることを明かとにした。すなわちTNF-α、IL-1β投与によってLCのPrP発現が増強し、GM-CSFによってPrPc発現が減少した。またPrP遺伝子欠損マウスを用いた検討では、LCにPrP発現は認められないことを報告した。以上から高純度で精製した表皮LCがPrP蛋白を発現していること、特定のサイトカインによってPrP発現調節の制御を受けることが示された。次年度では末梢血樹状細胞におけるPrP発現調節について検討する予定である。
