研究概要	昨年に引き続き幼若メラノサイトの分化に及ぼす因子を検討した。マウス神経冠培養系(primary culture)とマウス神経冠細胞由来メラノサイト前駆細胞株を使用し検討した。このメラノサイト前駆細胞株は当教室で樹立したcell lineで、NCCmelb4とNCCmelb5の2種ある前者はtyrosinase(TYR)陰性、DOPA反応陰性、endothelin B receptor(EDNBR)陰性の分化段階に留まる幼若な細胞であるため、メラノサイトの分化誘導因子を検討するのに最適である。また後者は全て陽性の発達のすすんだ段階の細胞で、コントロールとして使用した。悪性腫瘍の分化、転移に関して検索されている弾力線維の成分であるエラスチンのレセプターがNCCmelb4に発現していることをみつけ、エラスチンが未分化メラノサイトの分化に関連しているかを検討した。分化は電顕でメラノソームを観察し、光顕下で樹枝状突起の長さを測定した。またAramaar Blue Assayで細胞数を数えた。エラスチンはprimary cultureのメラノサイトおよびNCCmelb4を分化させるが増殖はさせなかった。最近、悪性腫瘍株にエラスチンのレセプターが発現しているとの報告がある。細胞の増殖なしに分化を促進するエラスチンは、悪性黒色腫の治療にも有用となりうる可能性があると考える。
