精神分裂病関連遺伝子操作動物の作製とその行動および脳内分子の解析

研究概要

1.mrt-1遺伝子改変動物の作製
これまでの研究から、新規遺伝子mrt1のスプライシングバリアントのうち、mrt1bが覚せい剤によって発現が増加し、統合失調症モデルの逆耐性現象に関与することが明らかになっていることから、CaMKIIプロモーターを用いた前脳部特異的にmrt-1bを過剰発現させた遺伝子改変マウスを作成した。現在F1世代において、2系統のジャームライン・トランスミッションが確認されている段階である。
2.Mrt-1と相互作用を持つ蛋白質の検索
精神分裂病(統合失調症)様症状発現薬に発達に依存して特異的応答を示すMrt-1bと相互作用を持つ蛋白質を、酵母Two-Hybrid法を用いて検索した。ヒトMrt-1b全長をプローブにして、クロンテック社ヒト脳cDNAライブラリ(4x10〜8クローン)をクロンテック社Matchmaker3の標準的方法を用いてスクリーニングした。Mrt1bにHAタグを、得られたクローンにMycタグをつけて哺乳類培養細胞HEK293に遺伝子を導入し、これらのタグに対する抗体を用いて免疫共沈降による検証を行って、これまでに6つのMrt-1b結合蛋白質をクローニングした。うち2つが既知蛋白で、ひとつはphosphatase、もうひとつはSAMドメインおよびPIDドメインを持つ蛋白であった。4つは未知蛋白であった。また、各クローンともPDZ、PXを含むN末端を削ったMrt1bとの間では免疫共沈降を示さないことも確認された。
