染色体18番における内因性精神病疾患感受性遺伝子の同定

研究概要

本研究は、染色体18p11.2領域の内因性精神病責任座位の絞込みと最終的な疾患感受性遺伝子の同定とを目的としている。18p11.2領域は、我々が報告した細胞内情報伝達に関係する新規遺伝子IMPA2遺伝子を初めとして、精神疾患に対する危険因子(疾患易罹患性因子)が存在する可能性が高い領域として注目されている。昨年度は、18p11.2領域を280kbで網羅する高密度マーカーを用いて日本人統合失調症80家系240名、214人の統合失調症孤発患者と313人の健常対照者について解析し、IMPA2遺伝子の1.2Mb下流に存在するC18orf1遺伝子についてD18S852、6409T>Cで構成されるハプロタイプで非常に強い関連を示唆する所見を報告した(P=0.00001)。本年度は、まず全染色体について444マーカー(染色体18番については27マーカー)を用いて網羅的に連鎖不平衡マッピング・伝達連鎖不平衡テストを行い、11q13.3に新たな疾患感受性座位が存在する可能性があることを報告したが、18p11.2領域については関連を示唆する所見は得られなかった。しかし、IMPA2遺伝子およびその近傍に位置する17SNPsを用いて、496人の気分障害患者と543人の健常対照者について遺伝子関連研究を行ったところ、promoter領域に存在する-1051G>T、-708G>A、-461C>T、およびイントロン1に存在するIVS1+1801C>Tで構成される特定のhaplotypeと疾患との関連が示唆された(P=0.007)。
