副甲状腺ホルモンの骨形成促進作用とエストロゲンによるその増幅機序に関する研究

研究概要	昨年度の研究において、副甲状腺ホルモン(PTH)はTGFβ特異的SmadであるSmad3発現を促進し、このSmad3がPTHの抗apoptosis作用に関わっていることを示した。本年度の研究では、PTHはSmad3発現促進のみならずTGFβ誘導性のSmad3転写活性を増強する作用を有することをSmad3反応領域を含むluciferase assayにより示し、そしてこれによりTGFβ誘導性のコラーゲン合成を促進することを見出した。即ち、PTH-Smad3 axisは骨芽細胞において抗apoptosis作用とTGFβ誘導性骨形成促進作用の増強という骨形成シグナルとなり、PTHの骨形成促進機序に関与することを証明した。またPTH及びSmad3過剰発現は骨芽細胞のβ-catenin発現を促進する結果を得、PTHの骨形成促進機序にWnt-β-cateninシグナルの増強が関与する可能性を提起した。一方、骨芽細胞におけるPTHとエストロゲンの相互作用に関しては、エストロゲンはPTHの増殖抑制作用を阻害し、またPTHはエストロゲン存在下で骨芽細胞機能を促進することを示した。そしてこの機序にエストロゲンにより骨芽細胞から産生が促進されるIGF-IとIGF結合蛋白-5が関与することを見出した。以前我々はエストロゲンがPTHの破骨細胞分化促進作用を阻害することを示したが、本年度の研究より骨形成系においてもPTHとエストロゲンは相互作用を有し、これによりエストロゲンはPTHの骨形成促進作用を増強する作用を有することを見出した。本研究により、PTHの骨形成促進機序とエストロゲンによるその増幅機序の一端が解明された。
